Characterization of the biocontrol of Phytophthora infestans and Alternaria solani by bacteria isolated from Belgian agroecosystems

Abstract
Potato is a major crop in Belgium and large quantities of agrochemicals are used to manage potato late blight, caused by Phytophthora infestans, and early blight, due to Alternaria solani. The present study aims at taking benefit from bacteria isolated from agroecosystems in Belgium for the biological control of these diseases. A collection of bacteria belonging to the Bacillus and Pseudomonas genera was created and their antagonistic activities against P. infestans and A. solani were assessed in vitro using direct confrontation tests on liquid and solid media. The strain 30BB6 of Bacillus subtilis inhibited the growth of P. infestans and A. solani respectively of 60 and 55%. A preliminary genetic characterization indicated that the bacterium possesses some genes related to the production of antimicrobial compounds (e.g. lipopeptides and siderophores) but none of the six genes associated to known virulence factors for vertebrates were detected by PCR for this strain. In planta tests performed in greenhouse under controlled conditions, revealed that this bacterium protect potato plants against P. infestans and A. solani, respectively by 67 and 70%. The capacity of the strain 30BB6 to control the potato late blight was then evaluated in the field. A significant reduction of the late blight severity occurred with a protection index of 22% compared to untreated plants. Experiments performed under controlled conditions showed the systemicity of the protection conferred by the strain 30BB6. The metabolic pathways involved in the induced systemic resistance is now studied in the interaction Solanum lycopersicum – Alternaria solani.
