Before the availability of next-generation DNA sequencing technologies that initiated the microbiome revolution in the early 2000s, microbiologists had to use and improve the conditions of in vitro culture of bacteria, revealing the dependence of the microorganisms on the availability of adequate substrates to grow and survive.
