Abstract
Cryopreservation of prepubertal testicular tissue is the only option to preserve the fertility of young cancer patients. In vitro maturation of cryostored tissue is aimed at obtaining spermatozoa to use in in vitro fertilization. No culture system has been able to reproduce the entire spermatogenic process from prepubertal testicular tissue. We first developed an organotypic culture system to study the immature testicular niche. Testicular fragments collected from 3 patients aged 2, 11 and 12 years were cultured showing a good preservation of the seminiferous tubules integrity and functional Sertoli and Leydig cells. The expression of two proteins of the blood testis barrier was also demonstrated but no germ cell differentiation was obtained. Optimization of our culture system with different media to culture fragments of 5 patients aged 2 to 12 years led to the generation of haploid cells as a significant previously unreported achievement.
