Adenosine triphosphate and phosphorylcreatine breakdown in resting and stimulated muscles after treatment with 1-fluoro-2,4-dinitrobenzene

Abstract
Sartorius muscles of male Rana temporaria were poisoned with 1-fluoro-2,4-dinitrobenzene. The ATP content of the resting muscles decreases rapidly within 15 min, at a rate nearly equal to that expected if the normal resting usage of ATP goes on, and the ATP production is inhibited. After 35 min incubation in 0.41 mM 1-fluoro-2,4-dinitrobenzene at 0°C, a 10 sec isometric tetanus provokes the breakdown of 1.14 micromoles ATP and 1.54 micromoles phosphorylcreatine per gram muscle; a 20 sec tetanus doubles these values. After 60 min incubation in 0.82 mM 1-iluoro-2,4-dinitrobenzene, a 10 sec tetanus provokes the breakdown of 0.83 micromoles ATP per gram, but no breakdown of phosphorylcreatine. After such tetanus, the muscles relax completely and there is no resynthesis of ATP within 200 sec. The ATP used is not found as ADP or AMP, but is dephosphorylated and deaminated to IMP.
