Abstract
Glioblastoma (GBM) is the most aggressive brain tumor, and despite its standard of care therapy it always recurs. In this thesis, we aimed at improving the GemC12-LNC hydrogel formed by lipid nanocapsules loaded with lauroyl-gemcitabine (GemC12-LNC) using it as a scaffold for dual drug delivery and exploiting its nanomedicine properties to increase the therapeutic efficacy against GBM. We demonstrated that paclitaxel, curcumin and salinomycin could be encapsulated in the GemC12-LNC hydrogel with a high encapsulation efficiency and without altering its physicochemical properties. Salinomycin was selected because of the synergic interaction with GemC12 and its activity against glioma stem cells. Finally, the NFL peptide was included in the GemC12-LNC hydrogel to promote its uptake in GBM cells, obtaining an increased cytotoxicity in vitro and a prolonged animal survival. In conclusion, this work improves the potential of the GemC12-LNC hydrogel for the local treatment of GBM.
