(en) Neuroinflammation is a central feature of several neurodegenerative
diseases and contributes significantly to disease progression. Effective
therapeutic strategies remain limited. In recent years, extracellular vesicles (EV)
have emerged as innovative and promising drug delivery systems. In this context,
loading therapeutic molecules within EV may enhance drug delivery and
therapeutic efficacy. This thesis aimed to develop bioactive lipid- and
microRNA-loaded EV designed to target and alleviate neuroinflammation. To
this end, multiple aspects of EV loading were investigated, the choice of EV
source was critically evaluated, and the therapeutic potential of 15d-PGJ2-
and miR-181c-loaded EV was assessed. This work underscored both the
potential and the limitations of EV-mediated delivery of therapeutic molecules.
