Abstract
Introduction : Adipose mesenchymal stem cells (AMSCs) are a promising source of multipotent cells for regenerative medicine. While AMSCs from abdominal adipose tissue have been well-investigated, those from the supra-fascia lata region remain largely underexplored, although the deep fat layer of the lateral tigh area is a prominent donor site in humans. This study compares the differentiation potential of AMSCs from these two anatomical regions in a porcine model. By investigation of their similarities and differences, this research may offer valuable insights into how anatomical origin influences regenerative potential. These insights could help determine the most suitable tissue source for regenerative applications such as bone regeneration and cartilage repair. Materials and Methods: Adipose tissues were sterilely harvested from the abdominal and supra-fascia lata regions of Landrace pigs (n = 4). The tissues were minced and digested in 0.2% collagenase type 1-A at 37°C for one to two hours. The collagenase activity was stopped by adding the proliferation culture medium supplemented with fetal bovine serum, penicillin-streptomycin (1%), and amphotericin B (1%). The cells were washed, centrifuged at 1500 rpm, and resuspended in culture medium. After four passages, performed to enrich for proliferative populations, cells were characterized by immunohistofluorescence to identify the expression of CD105, CD90 and CD73, and then differentiated into osteogenic, adipogenic, and chondrogenic lineages. Results: The immunohistofluorescence analysis confirmed the expression of mesenchymal stem cell markers (CD105, CD90 and CD73) at passage four for both cell sources, indicating successful selection of stem cell. No significant differences were observed in the differentiation potential of the two sources into osteogenic, chondrogenic, and adipogenic lineages (p > 0.05). However, there was a slight superiority in the adipogenesis potential of the supra-fascia lata-derived cells, suggesting the presence of regional influences on differentiation. Conclusion: This study provides valuable insights into the differentiation potential of AMSCs from two distinct anatomical regions. Although no statically significant differences were noted in their differentiation abilities, a subtle tendency towards variation in adipogenesis implies possible regional influences on lineage commitment. These findings especially lend credence to the feasibility of bone regeneration applications involving AMSCs derived from the abdominal and supra-fascia lata regions. Future research should also consider additional factors such as age, sex, and lifestyle habits which could further refine the application of AMSCs in targeted tissue repair in humans.
