Abstract
This thesis aims to study the 3D organization and coordinated development of portal space components. First, using a tissue clearing method and light sheet microscopy, we described how periportal mesenchyme forms the hepatic arteries, in parallel with bile duct morphogenesis. Second, we analyzed the transcriptome of cholangiocytes and adjacent mesenchymal cells during late embryonic development. Our data suggest that axon guidance genes are candidate regulators of portal tract development. Using transgenic knockout mice, we inactivated selected genes in the developing mouse liver and found that Slit2/Robo2 signaling is required in the mesenchyme as a novel player in hepatic artery development.
