Abstract
In recent years, in vitro culture of ovarian tissue has emerged as a promising method to obtain mature metaphase II oocytes from the primordial follicle pool in cryopreserved ovarian cortex, avoiding the need for tissue transplantation in patients at high risk of malignant cell reintroduction. However, this technique remains highly challenging, with very low follicle viability and developmental success rates. The in vitro culture system that best resembles the intraovarian environment is expected to cause the least follicle distress. In this context, this thesis presents two innovative strategies to improve in vitro follicle outcomes: (1) culture at 5% oxygen tension and (2) supplementation of culture media with adipose tissue-derived stem cell-conditioned medium.
