Bioassays were carried out under controlled conditions to investigate on the ability of the whitefly Trialeurodes vaporariorum to transmit Pepino mosaic virus (PepMV), a potexvirus usually spread by mechanical contact in tomato glasshouses. The insects spread the PepMV during high rate insects’ infection using the variety Moneymaker tomato as source as well as target plants. To determine if the transmission was due to the external presence of PepMV on whiteflies, the insect’s bodies were washed and the washing buffer used for virus detection by RT-PCR. A very low rate of external presence of PepMV particles was pointed out. This result was confirmed with absence of sap accumulation on whiteflies body under binocular microscope and microscope, at the contrary of other insects on tomato. Altogether these results suggest a transmission linked to the nutrition activity of whiteflies or a not yet known transmission mode. Interestingly, all the virus species vectored by Trialeurodes vaporariorum have a flexuous filamentous particle. Also, some potexviruses have been reported as vectored by insects. This new finding raises the need to reconsider non-conventional virus mode of transmission in the context of quarantine plant viral diseases.
