Abstract
Endocannabinoids and related N-acylethanolamines (NAEs) are known for their anti-inflammatory properties. Therefore, this work aimed to assess the effects of inhibiting their metabolizing enzymes in the context of acute lung injury (ALI) and multiple sclerosis. The endocannabinoid system being rather large, we focused on the less studied hydrolytic enzymes of 2-AG and NAEs. ABHD6 (α/β hydrolase domain 6), one of the enzymes regulating 2-AG levels was assessed in ALI. In multiple sclerosis, we focused on the enzymes hydrolyzing the NAEs and compared the potential neuroprotective properties of NAAA (N-acylethanolamine acid amidase) inhibition, FAAH (fatty acid amide hydrolase) inhibition and the dual inhibition of both enzymes. During our work on lung inflammation, we were intrigued by the distinctive alterations of the levels of 25-hydroxycholesterol (25-OHC). This prompted us to study the impact of lung inflammation on 25-OHC metabolism as well as the effects of the administration of this lipid mediator on ALI and allergic asthma. Our results support the interest of studying the endocannabinoids and the oxysterols in inflammatory diseases.
