(en) [Role of canagliflozin (invokana®), a novel sglt2 inhibitor, in the management of type 2 Diabetes] The gliflozines are a new class of antihyperglycemic agents for Type 2 diabetes therapy. Their action is based on SGLT 2 (sodium-glucose cotransporter Type 2) inhibition, this transporter being responsible for tubular glucose reabsorption. This insulin-independent effect was shown to be associated with weight loss and reduction in blood pressure levels, particularly systolic blood pressure. Canagliflozin (Invokana®) has been the first gliflozin available in Belgium. The aim of this paper is to review recent scientific data dealing with the administration of canagliflozin in type 2 diabetic subjects in controlled trials (24-104 week duration). Invokana® is associated with an improvement of glycemic control versus placebo as well as with a statistical superiority (at least for canagliflozin 300 mg) when compared with sitagliptin or glimepiride. Side effects are mostly mycotic genital and urinary tract infections of mild severity. Signs of osmotic diuresis and/or volume depletion are also reported in a minority of patients. Hypoglycemic events may occur when Invokana® is combined with an insulin-secretory agent or insulin. Invokana® has now a place in the treatment strategy of type 2 diabetes, except in the presence of nephropathy (glomerular filtration rate lower than 60 ml/min/1.73 m2) which decreases its efficiency
