Abstract
Despite growing interest in cancer metabolism, knowledge of the metabolic control of tumor metastasis is still limited. Through the generation of cancer cell models characterized by increased invasive and metastatic potential, we identified a mitochondrial switch corresponding to an overload of the TCA cycle with preserved mitochondrial functions but increased mitochondrial superoxide production. This switch promoted metastasis through Src activation in mitochondria and induction of protein tyrosine kinase PTK2B/Pyk2. Consequently, targeting mitochondrial superoxide prevented metastasis in mouse models.
