Obesity is a known risk factor and driver of triple-negative breast cancer (TNBC) progression, as underlined by clinical associations and preclinical studies. The intestinal microbiota is now considered a key player in this context. Our main objective was to understand how obesity promotes the growth of TNBC. To this aim, we studied the contribution of a bacterial metabolite, acetate, to the metabolism of TNBC in vitro and in vivo. Next, we assessed whether acetate could reverse obesity-induced TNBC growth. Furthermore, we characterized the metabolic differences between TNBC tumors from lean and obese mice using 13C metabolite tracing experiments. Our data show that acetate is not a key player but that obesity strongly affects tumor activity. Finally, in perspective, we explored the effects of modulations of the intestinal microbiota on tumor growth in an obesity context.
