Abstract
The Ysc-Yop type III secretion system allows extracellular Yersina bacteria, adhering to eukaryotic cells, to inject Yop effector proteins in the cytosol of these target cells. The injection of these effectors involves two events : the crossing of the bacterial envelope by the Ysc apparatus, generally called “secretion”, and the passage though the eukaryotic cell membrane by a pore, generally called “translocation”. <BR> several Ysc proteins, forming the Ysc apparatus, display homology with proteins of the flagellar basal body. The flagellum and the type III secretion system are not independent. There is a real cross-talk between the two systems (publication 1). <BR> The YopB and YopD proteins are required for the translocation pore formation. Infection of macrophages, previously loaded by a fluorescent probe, by IcrV mutants engineered in a yopQ background doesn’t lead to pore formation. While these mutants are able to produce YopB and YopD. This indicates that LcrV is directly required for the translocation pore formation. The YscF-needles, without the help of YopB, YopD and LcrV, do not make pores into the membrane of nucleated cells. Pores can only be detected with multieffector mutants or with single yopN mutants indicating that translocating Yops obstruct the pore (publication 2) <BR> Additionally, we whow that GAP activity of YopE prevents the osmotic lysis of target cells that results from pore formation (unpublished results). <BR> The utilisation of the P. aeruginosa pack swarming phenomenon as a method to detect pore formation by Y. enterocolitica allows to confirm the results obtained in publications 2 (unpublished results). <BR> In annex of this work, the identification of substrates and chaperon from the Y. enterocolitica 1B Ysa type III secretion system, is presented (publication 3
