Abstract
Non-coding RNAs (ncRNAs) are key regulators of gene expression and protein function and play important roles in normal hematopoiesis and cancer. In chronic and acute myeloid leukemias (CML and AML), deregulation of microRNAs and long non-coding RNAs has been associated with leukemogenesis and treatment resistance. The aim of this thesis was to evaluate the therapeutic potential of ncRNAs in myeloid leukemias. To address this question, two complementary studies were performed, focusing on distinct ncRNA classes and therapeutic contexts in CML and AML. The first study examined the microRNA miR-92a-1-5p in CML, in the context of BCR::ABL-driven signaling and tyrosine kinase inhibition. This work aimed to determine whether modulation of miR-92a-1-5p influences leukemic cell survival and response to imatinib, thereby assessing its relevance as a therapeutic target in CML. The second study focused on the long non-coding RNA H19 in AML and addressed whether its deregulation contributes to resistance to the hypomethylating agent azacitidine. By assessing the functional impact of H19 modulation in experimental azacitidine-resistant AML models, this study sought to determine whether H19 represents a driver of drug resistance or a bystander of prolonged treatment exposure. Together, these two studies yielded contrasting outcomes, illustrating both the promise and the limitations of targeting ncRNAs for therapeutic purposes in myeloid leukemias.
