Abstract
The buccal route for drug administration has gained importance in the past years due to its accessibility and rapid onset of action. It has been described as a reliable alternative to the oral route, both for local and for systemic delivery of drugs, overcoming some of the drawbacks existent in oral administration, such as the harsh milieu along the gastrointestinal tract. To successfully develop active drugs, the prediction of their absorption profile across the buccal mucosa is of utmost importance. Due to the high costs, time-consuming nature, and ethical considerations of ex vivo and in vivo studies, in vitro models have been the first choice for the screening of the drugs' permeability. Among the several cell lines originating in the buccal mucosa of humans or animals, hamster cheek pouch cells, oral keratinocytes, and TR146 cells have been the most used for permeability studies. However, due to their primary nature, hamster cheek pouch cells can only be used for a low number of tests, which leads to a high variability in the results. Also, the cancerous origin of TR146 makes it less close to human buccal epithelium, as compared to oral keratinocytes derived from human sources, for permeability studies.
