Osteoarthritis (OA) is a multifactorial pathology defined by chronic degeneration of articular cartilage and subchondral bone. Hyaluronic acid, one of the major components of the extracellular matrix (ECM) of cartilage, is produced or degraded by hyaluronan synthases HAS1 and 2 and hyaluronidases Hyal1 and 2. This work studies the role of Spam1 hyaluronidase in the development of OA. In the knee of mice, we showed that the absence of Spam1 has a protective effect against spontaneous age-related and induced post-traumatic joint degradation. In vitro Spam1 KO chondrocytes expressed less Hyal 2 and Mmp13 metalloproteinase than control chondrocytes, whereas HAS2 expression was not significantly modified. Absence of Spam1 appears to protect the joint against cartilage and bone degradation and suggests this hyaluronidase, expressed by chondrocytes, plays a role in the pathogenesis of OA.
