Abstract
Thyroid embryogenesis requires reorganization of a non-polarized cell mass into a multitude of polarized spherical monolayers called follicles. During cancer establishment, the well-organized tissue transforms back into a non-polarized cell mass. Both processes involve multiple intercellular communication steps. In this work, we study the role of extracellular vesicles (EVs) produced by cells that mediate the transition between the cell mass and the spherical monolayers during thyroid follicle formation. We demonstrate that EVs produced by endothelial progenitor cells (EPCs) are capable of stimulating thyroid follicle formation in an ex vivo thyroid explants culture system and that the folliculogenic activity of EPC-EVs is dependent on an adequate basal lamina production by the endothelial cells. However, the mechanism by which the basal lamina influences the folliculogenic potential of EVs still needs to be elucidated. Finally, we started to study the implication of EVs in papillary cancer (PTC) establishment in vivo, using an inducible human PTC mouse model. We demonstrate that PTCs produce EVs and that those EVs can be harvested from PTCs after gentle tissue dissociation.
