Abstract
As there is an unmet medical need for the treatment of glioblastoma, the aim of this project is to prevent local tumor recurrences with an anticancer drug-loaded hydrogel directly injected in the tumor resection cavity. In this project, a photopolymerizable hydrogel loaded with paclitaxel nanoparticles was developed and the antitumor efficiency in glioma orthotopic mouse model was investigated. Then the hydrogel was loaded with paclitaxel nanoparticles and free temozolomide. The drug combination achieved synergistic effect in vitro and superior antitumor efficacy in vivo. In addition, the beneficial antitumor efficacy of combination treatments with paclitaxel nanoparticle-loaded hydrogel and oral administration of temozolomide was also demonstrated. In conclusion, hydrogel-based local drug delivery has potential to be included in the standard of care of glioblastoma.
