Abstract
Ras GTPases operate as key molecular switch that convey extracellular signals from surface receptors to the interior of the cell, thereby regulating essential processes like proliferation, differentiation and survival. Ras is ditributed in different types of plasma membrane microdomains and endomembranes, generating intracellular signals in all of these domains. One of the many known substrates of Ras is the MAP kinases ERK1/2. Spatial regulation of ERK1/2 MAP kinases is an essential yet largely unveiled mechanism for ensuring the fidelity and specificity of their signals. We have approached this question by selectively targeting H-Ras and ERK proteins to different subcellular locations: bulk membrane, lipid rafts, endoplasmic reticulum (ER) and the Golgi complex. In this study we have investigated how proliferation and differentiation stimulae, can differentialy activate H-Ras and ERK in a time and space regulated fashion.
