Acidosis and hypoxia of the tumor microenvironment can have a profound impact on cancer progression and treatment efficacy. Our objective was to evaluate the effects of different pharmacological treatments on tumor extracellular pH (pHe) and pO2, using magnetic resonance techniques, such as 31P-NMR spectroscopy, Chemical Exchange Saturation Transfer MRI and electron paramagnetic resonance. The effects on tumor pHe and pO2 of two different pharmacological treatments were studied: UK-5099, a mitochondrial pyruvate transporter inhibitor, and syrosingopine, a monocarboxylate transporter inhibitor. Our results show that monitoring the evolution of parameters involved in tumor progression, such as pHe and pO2, is possible in vivo using advanced magnetic resonance technologies.
