(en) Since a decade, numerous investigations have highlighted the anti-inflammatory potential of the phenolic compounds (PCs) found in pomegranate (Punica granatum L.), and more especially of ellagitannins, which are mainly located in fruit husks. This study aimed at evaluating the anti-inflammatory properties at the intestinal level of a phenolic extract from pomegranate fruit husks (the PomH extract), left as by-products of the juice industry, in order to determine a potential role in the prevention against inflammatory bowel diseases (IBDs). In a first part of the work, a review has highlighted the potential of pomegranate husk as natural prevention against IBDs. Since the bioaccessibility should be taken into account when evaluating the physiological effects of ingested compounds at the intestinal level, and since several in vitro digestion protocols have been described in the literature, with a wide range of experimental conditions, the first experimental part focused on the establishment of a simple, well-defined protocol of human in vitro pre-colonic digestion. In a second experimental part, the bioaccessibility of the PomH extract was evaluated through the use of the optimized in vitro model. In the third experimental part, the anti-inflammatory effects of the PomH extract were investigated by using Caco-2 cells, an in vitro model of human intestinal epithelium, submitted or not to inflammatory inducers. After 24h incubation, 3 pro-inflammatory markers, i.e. interleukin 6 and 8, and monocyte chemoattractant protein 1, were assayed both at their gene transcription and secretion levels. The PomH extract had an anti-inflammatory impact, both at the gene transcription and protein levels, the later phenomenon being possibly due to a direct molecular trapping. Taken as a whole, this work provides useful information concerning the health-promoting potential of pomegranate PCs in IBDs.
