Despite progress in the management of patients with squamous cell carcinoma of the head and neck (SCCHN), the mortality rate of patients with advanced disease remains high. Therefore, additional treatment options to improve the overall survival of these patients are needed. We showed that ribociclib, a cyclin-dependent kinase 4/6 inhibitor, has anti-tumoural activity in preclinical human papillomavirus-negative SCCHN models. However, ribociclib has only a cytostatic effect, suggesting that its activity has to be optimized in combination with other treatments. Given that epidermal growth factor receptor (EGFR) overexpression is observed in 90% of SCCHN, we hypothesized that the combination of cetuximab, an antibody targeting EGFR, and ribociclib would be beneficial for patients with advanced SCCHN. However, we found that cetuximab was not the ideal partner to combine with ribociclib in all cases.
