Abstract
Metabolic dysfunction-associated steatohepatitis (MASH) is the progressive form of fatty liver disease and a recognized cardiovascular risk factor (CVD). However, the mechanisms linking hepatic pathology to CVD remain poorly understood. Adverse cardiac and vascular alterations accompanying steatohepatitis were characterized in two MASH mouse models: Foz mice on a high fat diet and C57BL/6JRj fed a western diet with fructose Both models exhibited cardiac fibrosis and dysregulation of genes involved in pathological remodelling, as well as impaired •NO-dependent vasorelaxation without atherosclerotic lesions. Our findings reveal that MASH is consistently associated with adverse structural but not functional alterations in the heart, with ADMA emerging as a key liver-derived mediator of endothelial dysfunction.
