initiation and the maturation of glandular trichome development. Trichomes are uni- or multicellular epidermal cell outgrowths covering the aerial organs of most plant species. Among those, glandular trichomes, characterized by a gland at the tip of the trichome secreting or storing secondary metabolites are highly organized structures suggesting the existence of molecular processes controlling different steps of their formation. Our understanding of control of their development is not as advanced as the one of unicellular non-glandular trichomes such as those found in Arabidopsis thaliana. Ectopic expression of AmMIXTA, a R2R3 MYB transcription factor in Antirrhinum majus can induce glandular trichome formation in tobacco, indicating that it acts as a genetic “master regulator”. Aiming to unravelling the molecular mechanisms driving early steps of glandular trichome development in tobacco, we carried out an RNA-seq study on transgenic Nicotiana tabacum lines expressing AmMIXTA under control of a -estradiol inducible promoter at different time points (4h and 48h). The identification of a pool of genes significantly upregulated following AmMIXTA induction enriched in regulatory genes, like transcription factors typically involved in trichome developmental processes (bHLH, HD-ZIP,C2H2 zinc finger MYB, ..), provides new insights on this developmental mechanism.
