Abstract
Despite advances in the treatment of patients with squamous cell carcinoma of the head and neck, the mortality rate in advanced and/or metastatic patients remains high. A better understanding of the resistance mechanisms involved could therefore be beneficial in improving treatment response. In this thesis, through two different window-of-opportunity studies, we analyzed patient samples collected before and after treatment with afatinib (EORTC-90111 study) and therapeutic vaccine (HN1901 study). The clinical and biological effects of the drugs were investigated, giving insights in cancer biology in response to a short preoperative anti-cancer therapy regimen. Hypotheses were raised concerning potential resistance mechanisms, as well as combinations of therapies that could have a synergistic effect.
