Abstract
The Amyloid Precursor Protein (APP) is implicated in the production of the amyloïd peptide responsible for the formation of senile plaques found in Alzheimer disease brains. Beside this toxic function, APP plays an important role in the maintenance of neuronal activity by mecanisms not yet fully understood. In this work, we studied particularly the role of APP in transcriptional regulation of genes implicated in neuronal activity. We observed that the expression of Npas4 is downregulated in the absence of APP. This alteration induces an increased expression of inhibitory (I) transmission markers and affects the excitatory (E) neurotransmission suggesting that the maintenance of the E/I neuronal equilibrium may be controlled by the APP through the expression of Npas4. Our data provide new insight to understand the complex role of APP in neuronal activity but also a mechanistic frame to further explore the impairments of network activity in Alzheimer’s disease.
