Nutrient Insulin Secretagogues Decrease Ca-45(2+) Efflux From Islet Cells By a Mechanism Other Than the Inhibition of the Na+-ca2+ Countertransport

Abstract
<jats:p>The mechanism whereby nutrient insulin secretagogues decrease <jats:sup>45</jats:sup>Ca<jats:sup>2+</jats:sup> efflux from islet cells is controversial. It was studied with mouse islets perifused with Ca<jats:sup>2+</jats:sup>‐free solutions. In the presence of Na<jats:sup>+</jats:sup>, glucose and ketoisocaproate inhibited <jats:sup>45</jats:sup>Ca<jats:sup>2+</jats:sup> efflux by about 50%. Substitution of choline<jats:sup>+</jats:sup> salts for Na<jats:sup>+</jats:sup> salts decreased the efflux rate by 45%, but did not prevent glucose from decreasing it further. Ketoisocaproate also inhibited <jats:sup>45</jats:sup>Ca<jats:sup>2+</jats:sup> efflux, but less markedly than in an Na<jats:sup>+</jats:sup> medium. Omission of Na<jats:sup>+</jats:sup> decreased the efflux rate even when it was already lowered by glucose or ketoisocaproate. It is thus clear that nutrient insulin secretagogues decrease <jats:sup>45</jats:sup>Ca<jats:sup>2+</jats:sup> efflux from islet cells by a mechanism other than the inhibition of the Na<jats:sup>+</jats:sup>‐Ca<jats:sup>2+</jats:sup> countertransport, possibly by increasing sequestration of the ion in cellular organelles.</jats:p>
